21 Claims

1. A method comprising:

translocating an expanded polynucleotide through a nanopore;

controlling the translocation of the expanded polynucleotide through the nanopore in a ratcheted manner; and

taking one or more electrical measurements as the expanded polynucleotide translocates through the nanopore.
