1. A compound of Formula (I), or a tautomer or a pharmaceutically acceptable salt thereof,

R is L-R1;
L is CH2, O, or SO2;
R1 is optionally substituted aryl or optionally substituted heteroaryl;
each R2 is independently H or alkyl;
R3 is optionally substituted alkyl, optionally substituted alkylene-OR2, optionally substituted cycloalkylene-OR2, optionally substituted alkylene-N(R7)2, optionally substituted cycloalkylene-N(R7)2, optionally substituted alkylene-C(O)N(R2)2, optionally substituted cycloalkylene-C(O)N(R2)2, optionally substituted alkylene-S(O)2N(R2)2, or optionally substituted cycloalkylene-S(O)2N(R2)2;
R4 is alkyl, optionally substituted alkylene-aryl, or optionally substituted alkylene-heteroaryl;
each R7 is independently selected from the group consisting of H, alkyl, (alkyl)C(O)—, (aryl)C(O)—, (alkyl)S(O)2—, and (aryl)S(O)2—;
X is —C(O)—, CH2, CHR6, or C(R6)2;
each R6 is independently selected from the group consisting of H, alkyl, and optionally substituted alkylene-OH;
X′ is —C(O)—, CH2, CHR3′, or C(R3′)2; and
each R3′ is independently selected from the group consisting of optionally substituted alkyl, optionally substituted alkylene-OR2, optionally substituted cycloalkylene-OR2, optionally substituted alkylene-N(R7)2, optionally substituted cycloalkylene-N(R7)2, optionally substituted alkylene-C(O)N(R2)2, optionally substituted cycloalkylene-C(O)N(R2)2, optionally substituted alkylene-S(O)2N(R2)2, and optionally substituted cycloalkylene-S(O)2N(R2)2;
provided that X and X′ are not both —C(O)—.
