A fusion protein comprising a first fragment of a nCas9 protein, a chimeric insertion polypetide and a second fragment of the nCas9 protein from N-terminus to C-terminus, wherein the chimeric insertion polypetide is TadA-TadA*, wherein the TadA-TadA* is a polypeptide encoded by the nucleotide sequence shown in SEQ ID NO: 11; wherein
the first fragment of the nCas9 protein has an amino acid sequence as set forth in SEQ ID NO: 1; and
the second fragment of the nCas9 protein has an amino acid sequence as set forth in SEQ ID NO: 2.
