10. A method of preparing two or more dry reactant spots for use with a liquid sample, the method comprising:
drying a first reactant in liquid phase onto a first membrane substrate to form a first dry reactant spot, the first reactant comprising a first binding partner conjugated to a first dye; and
drying a second reactant in liquid phase onto a substrate selected from the group consisting of the first membrane substrate, a second membrane substrate, a bead, and an inside surface of the reaction vessel to form a second dry reactant spot, the second reactant comprising a second binding partner conjugated to a second dye, wherein at least one of the first dye and second dye is a polymer dye, wherein the first dry reactant spot and the second dry reactant spot do not overlap, and wherein the first dry reactant spot and second dry reactant spot are capable of forming a liquid reaction mixture comprising the first binding partner and the second binding partner when contacted with the liquid sample; and
placing the first membrane substrate inside a reaction vessel.
