1. A method of forming a self-amplifying mRNA/lipid nanoparticle (SAM/LNP) formulation, comprising mixing first a first fluid stream containing self-amplifying mRNA molecules and second fluid stream containing a mixture of lipids under conditions such that at least a portion of the lipids of the second fluid stream form lipid nanoparticles encapsulating at least a portion of the self-amplifying molecules of the first fluid stream, wherein the first fluid or second streams further include a reducing agent.
