1. A method of preparing a polypeptide for analysis, comprising: a) cleaving the polypeptide in a sample in a first digestion, wherein the cleaving produces at least two fragments of the polypeptide, b) cleaving at least one of the at least two fragments of the polypeptide of a) with neutrophil elastase; and c) analyzing the sample after cleaving the at least two fragments of the polypeptide with the neutrophil elastase.
