1. A method of characterizing polynucleotide molecules, said method comprising the steps of:
(a) providing a solid support comprising a surface;
(b) providing a plurality of polynucleotide molecules attached to the surface, wherein each polynucleotide molecule has a random coil state and comprises a concatemer of multiple copies of one or more target sequences,
wherein each polynucleotide molecule is attached to the surface within a binding region substantially equivalent to a projection of the random coil on the surface,
wherein the polynucleotide molecules are arranged on the surface such that the polynucleotide molecules can be resolved by optics of an imaging system; and then
(c) detecting complete or partial nucleotide sequences of said one or more target sequences from said polynucleotide molecules from images of said plurality of polynucleotide molecules obtained using said imaging system;
wherein the solid support is not a bead.
