1. A microfluidic device for processing a biological sample, comprising:
a first channel configured to receive a solution including said biological sample, said first channel having i) a first end coupled to an inlet port of the microfluidic device, and ii) a dead end;
a chamber in fluid communication with said first channel, wherein said chamber is configured to receive at least a portion of said solution from said first channel, and
a film sealing at least one of said first channel and said chamber, wherein said film is configured to employ different permeability characteristics under different applied pressure differentials.
