1. A system, comprising;
a first region of DNA that encodes:
(i) a first fusion protein, wherein the first fusion protein comprises a substrate recognition domain coupled to a DNA-binding protein;
(ii) a second fusion protein, wherein the second fusion protein comprises a substrate domain coupled to a subunit of RNA polymerase;
(iii) a protein kinase;
and
(iv) a protein phosphatase; and
a second region of DNA that encodes:
(i) an operator for the DNA-binding protein;
(ii) a binding site for the RNA polymerase operatively coupled to the operator for the DNA-binding protein, wherein the subunit of the RNA polymerase is configured to recruit the RNA polymerase to the binding site; and
(iii) a reporter gene encoding a reporter protein, wherein transcription of the reporter gene is initiated upon (1) binding the DNA-binding protein to the operator and (2) binding the RNA polymerase to the RNA polymerase binding site.
