1. A method of making a tobacco plant, the method comprising:
(a) inducing mutagenesis in a tobacco cell to produce a mutagenized tobacco cell, wherein the mutagenized tobacco cell comprises a mutation in an endogenous nucleic acid molecule encoding a PR50 polypeptide having the amino acid sequence of SEQ ID NO: 3; and
(b) obtaining the tobacco plant from the mutagenized tobacco cell, wherein the tobacco plant comprises the mutation, wherein the tobacco plant exhibits a reduced amount of nicotine relative to a corresponding control tobacco plant lacking the mutation, and wherein the tobacco plant exhibits reduced activity of the PR50 polypeptide as compared to a control tobacco plant lacking the mutation.
