1. A non-naturally occurring DNA-binding polypeptide comprising from N-terminus to C-terminus:
an N-terminus region consisting of residues N+115 to N+1 of a Xanthomonas Transcription Activator-like Effector (TALE) protein;
a plurality of Xanthomonas TALE-repeat units, the TALE repeat units comprising a repeat variable di-residue (RVD);
a C-terminus region of the TALE protein; and
a cleavage domain or a half-cleavage domain conjugated to the C-terminus region,
wherein the TALE repeat units are ordered from N-terminus to C-terminus to specifically bind to a target nucleic acid in genomic DNA.
