1. A method comprising:
(a) providing a first mixture comprising a single-stranded ribonucleic acid (ssRNA) and a double-stranded deoxyribonucleic acid (ds-DNA);
(b) reverse transcribing the ssRNA in the first mixture, wherein the reverse transcribing comprises labeling the ssRNA with a first molecular barcode to produce a barcoded double- stranded complementary DNA (ds-cDNA), thereby obtaining a second mixture comprising the ds-DNA and the barcoded ds-cDNA; and
(c) bringing the second mixture or derivative thereof in contact with a plurality of adaptors comprising a second molecular barcode, to couple adaptors of the plurality of adaptors to the ds-DNA and the barcoded ds-cDNA.
