1. A method of separating a mixture in an installation comprising:
a first system comprising a plurality of chromatography columns, an outlet line for collecting a raffinate and an outlet line for collecting an extract;
at least one second system placed downstream of the first system; and
at least one tank fed by one of said outlet lines of the first system and feeding the second system with a continuous stream;
the method successively comprising, in a cyclic manner, in a given part of the first system:
a step of collecting a raffinate, a step of injecting the mixture to be separated, a step of collecting an extract, and a step of injecting a mobile phase;
wherein the method further comprises the measurement of the purity and/or of the yield of at least one collected fraction chosen from the extract and the raffinate, said measurement of the purity and/or of the yield comprising the following steps:
the determination of the concentration of at least two species of the mixture to be separated in the fraction collected in the tank;
the determination of the purity and/or of the yield of at least one species of the fraction collected in the tank.
