1. A method for generating nucleic acid barcode molecules, comprising:
(a) providing a plurality of nucleic acid molecules coupled to a support;
(b) coupling one or more additional nucleic acid molecules to nucleic acid molecules of the plurality of nucleic acid molecules coupled to the support, thereby assembling a plurality of nucleic acid barcode molecules coupled to the support, wherein the plurality of nucleic acid barcode molecules comprises a plurality of target products and a plurality of off-target products; and
(c) digesting off-target products of the plurality of off-target products with an exonuclease, wherein target products of the plurality of target products comprise an exonuclease-resistant bond, and wherein fewer than 50% of the target products comprising the exonuclease-resistant bond are digested with the exonuclease.
