1. A method for identifying a genotype of recombinant inbred lines (RILs) in soybeans with pod-shattering trait, comprising steps of:
(1) mapping soybean anti-pod-shattering major quantitative trait locus (QTL) using two soybean parent plants and resulting RILs;
(2) performing PCR amplification using genomic DNAs of the two soybean parent plants and the resulting RILs as templates and with
a forward primer comprises a nucleotide sequence as shown in SEQ ID NO. 1 and
a reverse primer comprises a nucleotide sequence as shown in SEQ ID NO. 2;
(3) digesting PCR fragments generated in step (2) with HpaII to produce digested fragments; and
(4) determining the genotype of RILs based on presence of target digested fragments in the digested fragments in step (3);
wherein the major QTL in step (1) is mapped on chromosome 5 of the soybeans.
